Proline is a non-essential amino acid that plays a significant role in the structural system of the body, particularly in the synthesis and maintenance of collagen. Collagen is the most abundant protein in connective tissues such as skin, bones, tendons, and ligaments. Proline, along with other amino acids like glycine and hydroxyproline, is a major component of the collagen triple-helix structure. Scientific studies have demonstrated that proline is necessary for the stability and strength of collagen fibers (Kishimoto et al., 1998). Deficiency in proline or impaired proline metabolism can negatively affect collagen synthesis, potentially leading to weaker connective tissues. Additionally, some research suggests that proline supplementation may support wound healing and tissue repair, although human clinical evidence is limited.
While there is a clear biochemical basis for the requirement of proline in collagen production and, by extension, the structural integrity of connective tissues, robust clinical trials directly linking oral proline supplementation to improved musculoskeletal health outcomes are sparse. Therefore, while the scientific rationale exists, further research is needed to confirm the efficacy of supplemental proline for structural system health in humans.
