Phlorizin is a natural compound found in apple tree bark and some other plants and is primarily known for its ability to inhibit sodium-glucose co-transporters (SGLT1 and SGLT2) in the kidneys. This mechanism prevents the reabsorption of glucose in the proximal renal tubules, leading to increased urinary glucose excretion—a process termed glycosuria. Historically, phlorizin was used in research to induce glycosuria in animal models to study diabetes and renal physiology.

The discovery of phlorizin’s mechanism directly inspired the development of modern SGLT2 inhibitors (like canagliflozin, dapagliflozin, and empagliflozin), which are now approved medications for type 2 diabetes and have shown significant benefits for renal and cardiovascular health. While phlorizin itself is not used therapeutically due to poor oral bioavailability and gastrointestinal side effects, its scientific validation as a prototype SGLT inhibitor is robust.
