Agnusides are iridoid glycosides found primarily in the fruit of Vitex agnus-castus, also known as chaste tree or chasteberry. Traditionally, Vitex agnus-castus has been used in European herbal medicine for centuries to support female reproductive health, including the regulation of menstrual cycles, the alleviation of premenstrual syndrome (PMS), and support of uterine function. Historical texts and herbal compendia frequently mention its use for conditions believed to be related to hormonal imbalances, including amenorrhea and dysmenorrhea. The rationale is that compounds like agnusides may influence pituitary hormones, thereby affecting progesterone and estrogen levels indirectly, which can have downstream effects on the uterus. However, while the traditional use is well documented, scientific evidence specifically linking agnusides themselves to direct uterine support is limited. Most modern research on Vitex agnus-castus as a whole suggests some efficacy in managing PMS and mastalgia, but robust clinical trials focusing on uterine health per se are lacking. The evidence that does exist often centers on the whole extract rather than isolated agnusides. Therefore, the support for using agnusides for the uterus is largely traditional, with some indirect support from broader studies on Vitex agnus-castus.
