Guaranine, which is chemically identical to caffeine and extracted from the seeds of the guarana plant (Paullinia cupana), is used in many weight loss supplements. The scientific rationale for its use is primarily based on its stimulant properties, which are known to increase metabolic rate and promote thermogenesis—processes that can potentially support weight loss. Several clinical studies have shown that caffeine, including caffeine derived from guarana, can modestly increase energy expenditure and fat oxidation in the short term. For example, randomized controlled trials have demonstrated that caffeine-containing products can lead to small but statistically significant reductions in body weight when compared to placebo, especially when combined with other stimulants or in the context of calorie restriction.

However, the overall quality of evidence is moderate at best. Most studies are short-term, sample sizes are often small, and the observed effects on weight loss are generally modest (often less than 2 kg over several weeks to months). There is also variability in individual response, and potential side effects such as jitteriness, insomnia, and increased heart rate must be considered. While guaranine is a traditional component in Amazonian cultures, its use for weight loss is scientifically validated only to a limited extent, mostly due to its caffeine content.
