Evidence supporting the use of: β-nicotinamide mononucleotide (NMN)
For the health condition: Wrinkles
Synopsis
Source of validity: Scientific
Rating (out of 5): 2
β-Nicotinamide mononucleotide (NMN) is a precursor to nicotinamide adenine dinucleotide (NAD+), a molecule essential for cellular metabolism and energy production. Interest in NMN as an anti-aging and skin health supplement is rooted in its role in supporting NAD+ levels, which decline with age. Preclinical studies in animals have demonstrated that boosting NAD+ can improve various markers of aging, including some related to skin health. For instance, NAD+ is involved in DNA repair and cellular energy, both of which are important for maintaining youthful skin appearance and function.
However, direct evidence in humans specifically linking NMN supplementation to the reduction of wrinkles is limited. Some in vitro and animal research suggests that increasing NAD+ levels can enhance dermal fibroblast activity, support collagen production, and reduce oxidative stress—all factors that may contribute to fewer wrinkles. A small number of human studies have evaluated topical or oral NAD+ precursors for skin health, but robust, blinded clinical trials specifically assessing wrinkle reduction with NMN are lacking as of mid-2024.
In summary, while the biological rationale for NMN in supporting skin health and potentially reducing wrinkles is promising, the current level of direct scientific evidence in humans is still preliminary. Most claims are based on mechanistic studies or extrapolation from animal models, warranting a cautious interpretation until more rigorous human trials are completed.
