Saraca asoca, commonly known as the Ashoka tree (literally "sorrow-less"), is a plant belonging to the Detarioideae subfamily of the Fabaceae family β€” also referred to as the legume, pea, or bean family. The word Ashoka comes from Sanskrit, meaning "without sorrow" or "that does not cause grief." The accepted scientific name is Saraca asoca (Roxb.) W.J. de Wilde. It is sometimes incorrectly known as Saraca indica. Saraca indica Linnaeus is generally mistaken for Saraca asoca (Roxb.) de Wilde, the real Indian Ashok tree. The former, with bracteoles spreading on the pedicel, is said to belong to Siam, Malaya, Sumatra, and Java, whereas the latter, with erect bracteoles clasping the pedicel, is found only in Sri Lanka, India, and Bangladesh.

Synonyms recorded in the classical Ayurvedic literature include KaΓ±keli, Madhu puspa, Rakta pallava, Vanjulah, Hema puspa, Gatasoka, and Taamrapallava, among others.

1.2 Botanical Description

Saraca asoca is a rapidly growing tree species that can reach heights of up to 10–12 m. It prefers warm and humid climates and can thrive in various soil types, including clay, loam, and sandy soil. It is a handsome, small, erect evergreen tree with deep green leaves growing in dense clusters. Its flowering season is around February to April. The Ashoka flowers come in heavy, lush bunches and are bright orange-yellow in color, turning red before wilting.

The fruit of the Ashoka tree is pod-shaped, black, 10–25 cm long, and 4.5–5 cm wide, linear-oblong, tapering to both ends. Seeds are 4–8, ellipsoid-oblong, 3–8 cm long, and slightly compressed.

1.3 Geographic Distribution

The tree is native to the Indian subcontinent, with its native range extending from Pakistan to the west, through India and Bangladesh to Myanmar in the east, and from Bhutan and Nepal in the north to Sri Lanka in the south. Within its native range, it primarily grows in seasonally dry tropical forests. The Ashoka is a rain-forest tree. Its original distribution was in the central areas of the Deccan Plateau, as well as the middle section of the Western Ghats in the western coastal zone of the Indian subcontinent.

1.4 Plant Parts Used Medicinally

Plant parts used in Ayurveda include the inner bark (Ashoka chhal), flowers (Ashoka pushpa), and occasionally seeds for topical ointments. The stem bark of the tree is the principal constituent used for treating menorrhagia and genito-urinary disorders.

2.1 Ayurvedic Tradition

Saraca asoca has been widely distributed in India and has been one of the ancient identifiable medicinal plants in the Indian subcontinent and South Asian countries for thousands of years. Its medicinal value seems to have been recognized first in the Agnivesha Charaka Samhita, which dates back to about 1000 BCE. The Sushruta Samhita mentions its bark decoction to relieve uterine pain and manage irregular menstruation.

The importance of Ashoka for female genito-urinary problems and especially in menorrhagia was recognized around the ninth century of our era. Classical Ayurvedic texts classify it as a "friend of women" β€” it is known as the "friend of women" as it is specifically used in the management of raktpradara (menorrhagia) and other female reproductive problems.

In Ayurveda, its qualities (Guna) are described as light to digest and dry (Rooksha); its taste (Rasa) is astringent (Kashaya) and bitter; its post-digestive effect (Vipaka) is pungent; its potency (Veerya) is cold (Sheeta); and it balances Kapha and Pitta doshas.

2.2 Unani Medicine

Ashoka has been used in Unani Medicine (Tibb-e-Unani) and other traditional systems of medicine for a long time. Unani practitioners adopted Ashoka bark in preparations credited with cooling effects for menstrual conditions.

2.3 Cultural and Religious Significance

The tree is considered to be very spiritual and is often planted around Buddhist and Hindu temples. In Indian epic poetry, the Ashoka tree is mentioned in the Ramayana in reference to the Ashoka Vatika (garden of Ashoka trees) where Hanuman first meets Sita. The flower of the Ashoka tree is the state flower of the Indian state of Odisha.

2.4 Traditional Preparations

The plant has been prepared in a variety of traditional forms across medicinal traditions:

  • Ashokarishta: A traditional Ayurvedic fermented tonic (asava-arishta) crafted primarily from the bark of the Ashoka tree combined with supporting herbs, jaggery, and natural fermentation.
  • Asokadi Churna: A powdered mix for menstrual regulation. Ashokaghrita is another preparation, along with wine-based fermented preparations blending bark with jaggery and spices.
  • Decoction (Kadha): Bark boiled in water and consumed warm. Churna (powder): Taken with milk or honey.
  • Other parts β€” bark, flower, and seeds β€” have been used traditionally for the treatment of diseases and ailments such as skin diseases, haemorrhoids, wounds, diarrhoea, worms, ulcers, and bone fractures.

A study showed that a hot water extract of the bark administered to human adult females stimulates the uterus similar to ergot, but without producing tonic contraction.

2.5 Ethnobotanical Uses Beyond Gynecology

Ashoka has traditionally been employed to treat dysentery, colic, piles, biliousness, dyspepsia, and ulcers, and is also known to display CNS depressant activity and regulate irregular menstrual cycles. S. asoca has been used extensively in Ayurvedic and Unani medicine, particularly for the treatment of gynaecological conditions like leucorrhoea, uterine fibroids, menorrhagia, and dysmenorrhea. It has also been used in traditional medicine to treat infections, skin disorders, haemorrhoids, and urinary problems.

3. Key Constituents and Active Compounds

3.1 Overview of Phytochemistry

Many studies have documented various chemical compounds/constituents such as glycosides, flavonoids, tannins, steroids, and steroidal glycosides. The stem bark, leaves, and seeds of S. asoca are known to contain flavonoids, tannins, alkaloids, glycosides, polyphenolics, fatty acids, and saponins. The reported major phytoconstituents responsible for its therapeutic uses are epicatechin, procyanidin B2, lyoniside, nudiposide, catechin, leucocyanidin, and leucopelargonidin.

3.2 Bark Constituents

Wide varieties of natural compounds such as flavonoids, tannins, catechol, sterol, glycosides, (βˆ’)-epicatechin, leucocyanidin, leucopelargonidin, procyanidin P2, and 11β€²-deoxyprocyanidin B are present in the bark of the Ashoka plant. The stem bark has been found to yield triterpinoids including ursolic acid, lupeol, and glochidiol, as well as sterols including campesterol, Ξ²-sitosterol, and stigmasterol. Bark extracts also show the presence of leucopelargonidin, leucocyanidin, (βˆ’)-epicatechin, (+)-catechin, (βˆ’)-3β€²-deoxyepicatechin-3-O-Ξ²-D-glucopyranoside, lyoniside, (βˆ’)-epigallocatechin, and (βˆ’)-gallocatechin.

Alkaloids, carbohydrates, phenols, glycosides, quinones, coumarins, terpenoids, flavonoids, and tannins were present in bark, young stem, and leaf samples in a comparative phytochemical profiling study. The study also provided quantitative evaluation of gallic acid present in the bark, young stem, and leaves of S. asoca. The leaves were found to have the maximum content of gallic acid compared to bark and young stem.

3.3 Flower and Leaf Constituents

Leaves contain various carbohydrates, tannins, gallic acid, and ellagic acid. Flowers are rich in sarcasin, sarcadin, waxy substances, proteins, carbohydrates, and steroids. Seeds contain various fatty acids including oleic, linoleic, palmitic, and stearic acid.

3.4 Beta-Sitosterol

Ξ²-sitosterol is a naturally occurring compound with antioxidant, anticancer, anti-inflammatory, and immunomodulatory properties, and is an effective bioactive compound present in the bark, stem, and leaves of Saraca asoca.

3.5 Cardioprotective Compounds

The main bioactive constituents of Saraca asoca identified for protecting cardiac tissue by inhibiting the ACE enzyme include flavonoids, flavanols, catechins, anthocyanins, phenolic acids, polyphenols, tannins, resveratrol, polysaccharides, fiber, saponin, and sterols.

4. Mechanisms of Action

4.1 Uterotonic and Oxytocic Activity

The plant tones the uterus, stimulates oxytocic activity, and has antitumor activity. The plant is found to have spasmogenic, oxytocic, uterotonic, antibacterial, anti-implantation, antitumor, antiprogestational, and anti-oestrogenic activity. The therapeutic potential of Saraca asoca lies in its phytochemical constituents, including flavonoids, tannins, and saponins, which exhibit anti-inflammatory, astringent, and uterotonic properties.

4.2 Antioxidant Mechanisms

Using an in-vitro DPPH (1,1-diphenyl-2-picrylhydrazyl) model, investigators found that high phenolic content may be the reason for the antioxidant properties of different bark extracts of S. asoca. Gallic acid and ellagic acid can be found in the flower and bark, respectively, serving as potent antioxidant contributors.

4.3 Anti-inflammatory Mechanisms

Regular treatment of adjuvant-induced arthritic rats with Saraca asoca acetone extract at doses of 300 and 500 mg/kg for up to 21 days increased RBC and Hb, decreased WBC, ESR, and prostaglandin levels in the blood, and restored body weight, compared to control and standard (indomethacin) groups.

4.4 Anticancer Mechanisms

An in vitro study evaluated the cytotoxicity of crude methanolic bark extracts of Saraca indica on HeLa cell lines using the MTT assay. The extract possessed highly significant percentage of inhibition on the cell lines, and the IC50 value was determined to be 14.63 Β΅g/ml. These results indicate a significant antitumor and cytotoxic effect against human cervical cancer HeLa cell line.

4.5 ACE Inhibition and Cardiovascular Protection

Angiotensin I is broken down by the ACE-1 enzyme to Angiotensin II, which aggravates blood pressure by narrowing blood vessels. High ACE activity is detrimental to the heart by enhancing angiotensin II concentration. ACE inhibitors isolated from plant extracts are considered healing agents that inhibit the production of angiotensin II, thereby preventing hypertension and other cardio-related problems.

5. Scientific Evidence by Area of Use

5.1 Female Reproductive Health and Menstrual Disorders

This is the most extensively documented application of Ashoka, both historically and in modern research. Saraca asoca is known for its unique pharmacological role in treating various uterine complications, menstrual disorders, and bacterial infections, bleeding hemorrhoids, urinary and dermatological problems.

Dried Saraca asoca bark has been used for menorrhagia in India. Dried bark as well as the flowers are given as a tonic to women with uterine disorders. The stem bark is used to treat all disorders associated with the menstrual cycle. In Sri Lanka, the bark is used for menstrual disorders and menorrhagia.

Trials on Ashokarishta show it helps reduce heavy periods, balance cycle patterns, and boost general health in cases of menorrhagia or painful menstruation. An herbal uterine tonic derived from a variety of medicinal plant extracts was effective in treating various menstrual disorders such as abnormal bleeding, menorrhagia, irregular periods, and dysmenorrhoea.

Evidence strength: Ashokarishta is a traditional Ayurvedic fermented herbal tonic centered on Saraca asoca bark, used historically for women's reproductive wellness. While popular for menstrual comfort, scientific validation remains limited, so it should complement β€” not replace β€” evidence-based care. The existing clinical data derive mainly from small Indian trials, often without rigorous blinding or large sample sizes. More research is needed to systematically evaluate its phytochemical, pharmacological, and clinical properties, as well as to develop safety, pharmacology, and toxicology reports for traditional formulations.

5.2 Anti-inflammatory and Analgesic Activity

Laboratory investigations indicate potent antimicrobial, anti-inflammatory, antioxidant, antidiabetic, analgesic, cardioprotective, antiurolithiatic, anticancer, CNS depressant, and uterine tonic actions. In one experimental study, Saraca asoca extract showed significant antidiabetic and anti-inflammatory activities, with fresh and dry leaves exhibiting 75% and 25% efficacy respectively, with 60% antioxidant inhibition.

Evidence strength: Anti-inflammatory evidence is predominantly preclinical (animal models and in vitro). Robust human clinical trials specifically for inflammation or pain are absent from the published record.

5.3 Anticancer Activity

The anticancer principle from Saraca asoca flowers indicated 50% cytotoxicity (in vitro) in Dalton's lymphoma ascites and Sarcoma-180 tumour cells. Cancer-preventive experiments revealed that methanol bark extracts of Saraca indica can effectively diminish the cell growth of human cervical cancer cell lines (HeLa) in dose-dependent manners with a determined IC50 value of 14.63 Β΅g/ml.

More studies on quality control, standardization, and clinical trials on S. asoca should be performed, so that this plant could be utilized as an effective anti-cancer drug, which could be a promising source for the herbal and pharmaceutical industry.

Evidence strength: All anticancer evidence currently published is in vitro or animal-based. No human clinical trials have established Ashoka as a cancer treatment. Evidence at this stage is preliminary only.

5.4 Antidiabetic Activity

Saraca asoca is one of the leading medicinal herbs from ancient past and has several medicinal properties including antiviral, antidepressant, antimicrobial, antidiabetic, antiulcer, and anti-inflammatory, and anti-cancer activity.

Evidence strength: Antidiabetic evidence is based on in vitro assays and preclinical animal studies. No adequately powered human randomized controlled trials have been identified specifically for glycemic control.

5.5 Antimicrobial Activity

It has been used as an antibacterial agent and has antioxidant properties and antiviral properties. Ethanol and water extracts of flowers of Saraca asoca exhibited antibacterial activity against Staphylococcus aureus, Aeromonas liquefaciens, Trichophyton, Candida albicans, Klebsiella pneumoniae, and E. faecalis.

Evidence strength: Antimicrobial data are derived from in vitro studies. No human clinical trials have tested Ashoka extracts specifically as antimicrobial treatments.

5.6 Cardioprotective Activity

Cardioprotective activity of alcoholic extract of Saraca indica bark was investigated against cyclophosphamide-induced cardiotoxicity. Treatment with Saraca indica significantly (p<0.05) reversed the status of cardiac biomarkers, ECG changes, oxidative enzymes, and lipid profile in a rat model of cardiotoxicity.

Evidence strength: Cardioprotective evidence is limited to animal (in vivo) models. Human clinical evidence is absent.

5.7 Visceral Leishmaniasis

Indian Kala-azar (visceral leishmaniasis) is a protozoan parasitic disease caused by Leishmania sp. and is endemic in the Indian subcontinent. VL mainly targets people with deficiencies in protein, nutrients, and essential trace elements, which ultimately leads to immunodeficiency. A published PMC study explored the use of S. asoca seed extract in experimental murine visceral leishmaniasis, finding recovery of trace element imbalances β€” evidence that is currently preclinical only.

5.8 Menopausal Symptoms

A clinical trial found that the combined treatment of Ashokarishta, Ashwagandha Churna, and Praval Pishti resulted in a significant reduction in menopausal symptoms. This trial tested a combination formulation, making it difficult to isolate the specific contribution of S. asoca.

Evidence strength: One open-label multicenter study with a combination product. The attribution of efficacy to Ashoka specifically cannot be established from this design.

  • Urinary system: Saraca asoca has uses mainly in medicine to treat women's gynecological disorders, in all types of abnormal vaginal discharge, in uterine inertia, uterine pain, urinary calculus, and dysuria.
  • Skin and integumentary system: The bark of the Ashoka plant is used to prepare cosmetics that help to improve skin complexion.

7. Dosage Forms and Reported Dosages

Ashoka is available and used in several dosage forms. Dosages cited below are as reported in reviewed sources and should not be construed as prescriptive recommendations.

  • Bark powder (churna): Taken as a powder with milk or honey.
  • Decoction: Bark boiled in water and consumed warm.
  • Ashokarishta (fermented liquid): Prepared through natural fermentation of a herbal decoction with a sweetening medium (usually jaggery), resulting in a small amount of naturally occurring alcohol in the final product. The typical dosage cited by Ayurvedic practitioners varies from 15–30 ml, taken with an equal quantity of water, typically after meals. Specific dosages cited in published trials vary; Ashokarishta at 5 ml has been referenced in some sub-therapeutic evaluations.
  • Bark extract capsules/tablets: Several small clinical trials in India documented a dosage of 500 mg bark extract twice daily for menstrual pain assessment.

The concentration of (+)-epicatechin in processed drugs (Ashokarishta) from different manufacturers was found to vary significantly β€” as much as seven-fold between major brands β€” but no corresponding difference in antimicrobial activity was observed, indicating substantial variations among plant-based drugs. This highlights the importance of standardization.

8. Safety Considerations and Interactions

8.1 Pregnancy

Pregnant women should not take Ashokarishta. The safety profile of this medicine during pregnancy is not well-established. Given its documented uterotonic and oxytocic activity β€” historically exploited to stimulate uterine contractions β€” use during pregnancy carries a theoretical risk that warrants caution.

8.2 Preclinical Toxicology

A preclinical safety evaluation of aqueous bark extract of Saraca asoca (Roxb.) has been published, covering acute and subacute oral toxicity in animal models. More research is needed to systematically develop safety, pharmacology, and toxicology reports for traditional formulations. Formal human toxicological data remain limited.

8.3 Adulteration β€” A Major Safety Concern

The indiscriminate and rampant extraction of the wood to meet ever-increasing market demand has led to a sharp decline in naturally occurring populations of the species. Consequently, the species has recently been classified as "vulnerable" by the International Union for Conservation of Nature (IUCN).

The most common adulterant is the bark of Polyalthia longifolia Benth. & Hook (family Annonaceae), called "Khota ashoka" in Marathi and "Devdaru" in Hindi and Bengali. Ashoka bark is also mixed with bark of Rohitaka (Afanamexis polystachys) and Caesalpinia pulcherrima.

These results of widespread adulteration pose grave implications for the use of raw herbal drugs such as that of S. asoca on consumer health and safety. A strong and robust regulatory framework is needed to ensure the quality of raw herbal trade products and to reassure consumer confidence in indigenous medicinal systems.

8.4 Conservation Status and Supply Integrity

This species is classified as "vulnerable" under the IUCN Red List due to its dwindling population caused by destructive harvesting from natural habitats. Conservation and multiplication of this species is urgently needed to sustain its medicinal uses. The significant medicinal benefits of Saraca asoca have led to its premature felling, causing a sharp decline in the population. The rising demand has also led to an increase in product adulteration.

8.5 Evidence Gaps and Research Needs

The scientific data gathered in the course of research corroborate traditional claims about the health benefits associated with this plant and indicate the necessity for further clinical and pharmacological research to confirm its beneficial effects. More studies on quality control, standardization, and clinical trials on S. asoca should be performed so that this plant could be utilized as an effective anti-cancer drug, which could be a promising source for the herbal and pharmaceutical industry.
