Research suggests Ashwagandha's withanolides modulate the HPA axis, reduce cortisol and ACTH, and exhibit GABAergic activity that may account for its anxiolytic effects. For sleep, a PLOS ONE meta-analysis of 5 RCTs (n=400) found a small but significant improvement in overall sleep quality (SMD −0.59), with effects more prominent at doses ≥600 mg/day and treatment durations ≥8 weeks. Studies also demonstrate improvements in sleep onset latency, sleep maintenance (reduced wake after sleep onset), and mental alertness on rising. A systematic review confirmed that supplementation with 250–500 mg of Ashwagandha extract daily for 4 to 13 weeks significantly decreased morning cortisol levels in adults experiencing higher stress. L-theanine — 500 mg
L-theanine is a non-protein amino acid found naturally in tea (Camellia sinensis) that crosses the blood-brain barrier and promotes alpha-wave brain activity associated with calm alertness without sedation. A systematic review and meta-analysis of 19 articles (N=897 participants) found L-theanine significantly improved subjective sleep onset latency (p=0.04), daytime dysfunction (p<0.001), and overall subjective sleep quality (p=0.03). A single-dose study of 200 mg showed significantly increased frontal alpha-wave power and reduced salivary cortisol compared to placebo during an acute stress challenge. L-theanine also has antioxidant and anti-inflammatory properties, attributed to its ability to reduce reactive oxygen species synthesis and block the NF-κB signaling pathway.
Magnolia (Magnolia officinalis) (bark) extract standardized to 2% honokiol (6 mg) and 1% magnolol (3 mg) — 450 mg
Magnolia bark extract has been used for over 1,000 years in traditional Chinese and Japanese medicine for anxiety, nervousness, insomnia, and mood disorders. The first recorded mention appears in the Shennong Bencao Jing around 100 A.D. for treating emotional distress. Its active compounds — the biphenolic neolignans honokiol and magnolol — readily cross the blood-brain barrier and act as positive allosteric modulators of GABA-A receptors, providing anxiolytic effects consistent with benzodiazepine-site activity. In animal studies, the benzodiazepine-blocking drug flumazenil reversed magnolol's effects, confirming the specific receptor pathway. Honokiol also interacts with serotonin receptors (5-HT1B and 5-HT6) and dopamine receptors, consistent with its traditional calming and sleep-inducing uses. In a double-blind, placebo-controlled RCT (Talbott et al., 2013), a Relora® combination containing Magnolia (750 mg/day for 4 weeks) significantly reduced salivary cortisol, perceived stress, tension, and anger in moderately stressed adults versus placebo. A pilot RCT in premenopausal women showed reductions in state anxiety scores.
